ABSTRACT

OBJECTIVE To study the pharmacological basis of Schisandra chinensis in the treatment of allergic asthma. METHODS The common components of 10 batches of S. chinensis from different habitats were analyzed by UPLC-Q-TOF-MS/MS. Furthermore, the allergic asthma model was established by intraperitoneal injection of ovalbumin (OVA) and aluminum hydroxide for stimulation combined with atomization exitation; general behavioral observation and the contents of interferon γ (IFN-γ), interleukin-4 (IL-4) and immunoglobulin E (IgE) in serum were taken as criteria for evaluating the therapeutic effect of S. chinensis from different habitats in the treatment of allergic asthma. Correlation coefficients between common peak area and efficacy evaluation index of each batch of medicinal material were analyzed through grey correlation degree and Pearson correlation analysis. RESULTS A total of 21 common components were identified in 10 batches of S. chinensis from different habitats. After administration of S. chinensis, symptoms such as shortness of breath, sneezing and curling of rats were alleviated. In addition, the content of IFN-γ was significantly increased while the contents of IL-4 and IgE in serum were distinctly decreased (P<0.01). Grey correlation analysis showed that 11 common components had high correlation coefficients with IFN-γ, IL-4 and IgE (rˉ>0.8). Pearson correlation analysis showed that 8 components were significantly positively correlated with the content of IFN-γ (P< 0.05), and 9, 8 components were significantly negatively correlated with the content of IL-4 and IgE (P<0.05). Based on the results of grey correlation degree and Pearson correlation analysis, 7 components such as peak 3, 4, 6, 7, 9, 19 and 20, were highly related to S. chinensis in the treatment of allergic asthma. CONCLUSIONS Schisandrol A, schisandrin B, schisandrin C, gomisin M2, gomisin J, pregomisin and angeloylgomisin H are the potential pharmacodynamic substance basis of S. chinensis in the treatment of allergic asthma.

ABSTRACT

Objective:To establish the fingerprints of Plantaginis Herba.Methods:The fingerprints were determined by UPLC. The peak areas of fingerprints of different parts and origins were analyzed by variance analysis and independent sample t-test. PCA, HCA, PLS-DA and other chemical patterns were analyzed by Simca14.1. The index weight was calculated by CRITIC, and the quality of plantain evaluation was combined with grey correlation degree.Results:The fingerprints of grass, stem, leaf and spike of Plantago depressa Willd. calibrated for 24, 16, 23 and 22 common peaks. The fingerprints of grass, stem, leaf and spike of Plantaginis Herba calibrated for 22, 10, 16 and 22 common peaks, and the fingerprints of commercial mixed plantain calibrated for 23 common peaks. 10 peaks were identified. The analysis of variance showed that there were differences in chromatographic peak areas between different parts of Plantago asiatica L. and Plantago depressa Willd.. And combinedede with PLS-DA, it showed that there were 16 important characteristic indexes in the classification, and the importance ranking was peak 3, 8, 28, 12, 14, 7, 5, 17, 6, 19, 23, 11, 22, 27, 9, 16. The quality evaluation results of critical method combined with grey correlation degree showed that among Plantago depressa Willd., Plantago asiatica L. and commercial mixed plantain herbs, the quality of Plantago asiatica L. was the best. Conclusion:The mixture of plantain exists in the market. The fingerprints established in this study can be used for the identification and quality evaluation of Plantaginis Herba from different sources.

@#【Objective 】 To explore the influencing factors of Yang deficiency constitution in traditional Chinese medicine (TCM) from the perspective of mathematics with the use of calculation formulas, so as to protect patients from getting diseases caused by Yang deficiency constitution and provide suggestions for TCM disease prevention. 【Methods】  Based on the classification and determination criteria of TCM constitution implemented by China Association of Chinese Medicine, data for 24 solar terms from May 5, 2020(Start of Summer) to April 20, 2021 (Grain Rain) for the identification of Yang deficiency were collected by mobile constitution identification system. The grey correlation analysis method was used to determine the grey correlation degree of the factors influencing Yang deficiency constitution. In addition, a random forest model was constructed for the verification of the results from the grey correlation analysis, and for the evaluation of correlation degree between Yang deficiency constitution and its influencing factors. 【Results】  A total of 16 259 sets of data were collected from healthy or sub-healthy individuals aged from 18 to 60 years living in the central and northeastern parts of Sichuan Province(China) for the identification of TCM constitutions. After screening and preprocessing, a total of 544 sets of data for the identification of Yang deficiency constitution, involving 18 aspects of factors influencing Yang deficiency constitution. The results of the grey correlation analysis showed that there were 12 influencing factors whose grey correlation degree with Yang deficiency constitution was greater than 0.6. The accuracy of these 12 influencing factors with the training set and validation set of the Yang deficiency constitution random forest model were 98.39% and 93.12%, respectively. 【Conclusion】  In the sample data selected in this paper, grey correlation analysis is the appropriate technology to analyze the influencing factors of Yang deficiency constitution. It provides a new idea and a new methodological reference for the research and analysis of the influencing factors of TCM constitution.

ABSTRACT

The grey correlation-TOPSIS method was used to evaluate the quality of the origin herbs of Lonicerae Japonicae Flos, and the Fourier transform near-infrared(NIR) and mid-infrared(MIR) spectroscopy was applied to establish the identification model of origin herbs of Lonicerae Japonicae Flos by combining chemometrics and spectral fusion strategies. The content of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, secoxyloganin, isoquercitrin, isochlorogenic acid B, isochlorogenic acid A, and isochlorogenic acid C in six origin herbs of Lonicerae Japonicae Flos was determined by high-performance liquid chromatography(HPLC), and their quality was evaluated by the grey correlation-TOPSIS method. The Fourier transform NIR and MIR spectra of six origin herbs of Lonicerae Japonicae Flos(Lonicera japonica, L. macranthoides, L. hypoglauca, L. fulvotomentosa, L. confuse, and L. similis) were collected. At the same time, principal component analysis(PCA), support vector machine(SVM), and spectral data fusion technology were combined to determine the optimal identification method for the origin herbs of Lonicerae Japonicae Flos. There were differences in the quality of the origin herbs of Lonicerae Japonicae Flos. Specifically, there were significant differences between L. japonica and the other five origin herbs(P<0.01). The quality of L. similis was significantly different from that of L. fulvotomentosa, L. macranthoides, and L. hypoglauca(P=0.008, 0.027, 0.01), and there were also significant differences in the quality of L. hypoglauca and L. confuse(P=0.001). The PCA and SVM 2D models based on a single spectrum could not be used for the effective identification of the origin herbs of Lonicerae Japonicae Flos. The data fusion combined with the SVM model further improved the identification accuracy, and the identification accuracy of the mid-level data fusion reached 100%. Therefore, the grey correlation-TOPSIS method can be used to evaluate the quality of the origin herbs of Lonicerae Japonicae Flos. Based on the infrared spectral data fusion strategy and SVM chemometric model, it can accurately identify the origin herbs of Lonicerae Japonicae Flos, which can provide a new method for the origin identification of medicinal materials of Lonicerae Japonicae Flos.

ABSTRACT

ObjectiveHigh performance liquid chromatography (HPLC) was used to establish the specific chromatograms of Aurantii Fructus from different origins, and the quality variability of Aurantii Fructus from Sichuan was analyzed and evaluated by combining entropy weighting method and grey correlation method. MethodHPLC was performed on an Agilent Eclipse Plus C18 column (4.6 mm×250 mm, 5 μm) with a gradient elution of methanol (A)-0.1% phosphoric acid aqueous solution as the mobile phase (0-12 min, 25%-33%A; 12-21 min, 33%-41%A; 21-30 min, 41%-42%A; 30-40 min, 42%-59%A; 40-53 min, 59%-72%A; 53-60 min, 72%A; 60-65 min, 72%-100%A; 65-70 min, 100%A; 70~71 min, 100%-25%A; 71-80 min, 25% A) at a flow rate of 1.0 mL·min-1, the injection volume was 10 μL and the detection wavelength was 330 nm. Fifty batches of Aurantii Fructus samples from different origins (Sichuan, Chongqing, Jiangxi and Hunan) were tested, and the similarity evaluation software is used to generate characteristic profiles and compare them with control profile for peak identification, and then to evaluate the similarity of the samples. IBM SPSS 19.0 and SIMCA 14.1 were used to perform multivariate statistical analysis on the results of the samples, and then the entropy weighting method and grey correlation were used to calculate the overall quality score of samples from Sichuan. ResultHPLC specific chromatogram of Aurantii Fructus was established, and 14 common peaks were identified as eriocitrin, neoeriocitrin, narirutin, naringin, hesperidin, neohesperidin, meranzin hydrate, poncirin, meranzin, marmin, nobiletin, 3,3′,4′,5,6,7,8-heptamethoxyflavone, tangeretin and auraptene. And the similarities between the samples from Sichuan and the control chromatogram were all above 0.980. The samples could be classified into four categories according to their main origins by chemical pattern recognition, and the results of cluster analysis, principal component analysis and orthogonal partial least squares-discriminant analysis were all able to discriminate the samples of different main origins effectively. The comprehensive evaluation results of entropy weighting method combined with grey correlation showed that the quality of Aurantii Fructus from Sichuan varied greatly among different origins, and the quality of Aurantii Fructus from Sichuan was ranked as Bazhong>Luzhou>Chongqing>Neijiang. ConclusionIn this study, the characteristic mapping of Aurantii Fructus from Sichuan is established, and combined with the analytical methods of chemometrics and grey correlation, the quality of samples from different origins can be effectively differentiated, which can provide a reference for the comprehensive evaluation and control of the quality of Aurantii Fructus from Sichuan.

ABSTRACT

Objective:To explore the effects of different processed products of Epimedii Folium on cytotoxicity and the material basis of toxicity. Methods:By using the SRB method to investigate the effects of different processed products of Epimedii Folium on the proliferation of HaCaT cells; and based on the grey correlation analysis method to establish the data spectrum effect relationship of HPLC fingerprint spectrum-toxicity so as to determine the toxic components and processing methods. Results:The value of cytotoxicity IC 50 of different processed products of Epimedii Folium is as follow: vinegar fried> oil fried > original > single fried > salt fried > wine fried. Among the 12 characteristic chromatographic peaks, Peak No.3 (magnoflorine, correlation value: 0.870) and Peak No.6 (epimedin C, correlation value: 0.851) are highly correlated with the toxicity value. Conclusions:Both vinegar fried and oil fried Epimedii Folium have the effect of reducing toxicity. Magnoflorine and epimedin C may be the main toxic components in Epimedii Folium. The study provides scientific basis for the research on the process optimization of Epimedii Folium concocting to reduce toxicity.

ABSTRACT

OBJECTIVE To study the sp ectrum-effect relationship of anti-gastric ulcer effect of Shaoyao gancao decoction. METHODS Eleven batches of Shaoyao gancao decoction were prepared ;gastric ulcer model was established by anhydrous ethanol modeling method. Using Weikangling as positive control ,the effects of Shaoyao gancao decoction on the contents of defensive factors [nitric oxide (NO),epidermal growth factor (EGF),superoxide dismutase (SOD)] and attack factor [malondialdehyde (MDA)] in gastric tissue of model rats were investigated. HPLC fingerprints of 11 batches of Shaoyao gancao decoction were established and similarity evaluation was performed with Similarity Evaluation System of Traditional Chinese Medicine Chromatographic Fingerprint (2004A edition ); common peaks were identified by comparing with mixed control. The spectrum-effect relationship of Shaoyao gancao decoction against gastric ulcer was analyzed based on the grey correlation analysis. RESULTS Eleven batches of Shaoyao ganyao decoction could significantly decrease the content of MDA in gastric tissue ,while increased the contents of NO ,EGF and SOD in gastric ulcer model rats (P<0.01),and had a certain inhibitory effect on the gastric ulcer. There were 23 common peaks in chromatograms of 11 batches of samples ,and the similarity with the control fingerprint was not less than 0.9. By comparing with mixed control ,7 common peaks were identified ,namely gallic acid (peak 5),albiflorin(peak 9),paeoniflorin(peak 10),liquiritin(peak 12),isoliquiritin apioside (peak 14),isoliquiritoside(peak 15), glycyrrhizic acid (peak 22). The average correlation degree of 7 identified common peaks and pharmacodynamic indexes were greater than 0.6,of which peak 22(glycyrrhizic acid ),peak 10(paeoniflorin)and peak 12(liquiritin)had the largest correlation , and their values were 0.807,0.772 and 0.770 respectively. RESULTS The anti-gastric ulcer effect of Shaoyao gancao decoction is the result of the synergistic effect of multiple components ,among which glycyrrhizic acid ,paeoniflorin and liquiritin may be the main pharmacodynamic components.

ABSTRACT

Objective:To analyze the composition, the changes of expense structure and the influencing factors of hospitalization expenses, for reference in optimizing the cost control of day surgery.Methods:Collection of the first page data of patients with the top three diseases(varicose veins of lower limbs, chronic cholecystitis and varicocele)in the day surgery volume ranking in three tertiary general hospitals in a city in 2020. The confounding factors were eliminated through propensity matching. The structural change of hospitalization expenses was analyzed by structural change degree, and the influencing factors of hospitalization expenses were analyzed by grey correlation degree and multiple linear regression.Results:After 1∶1 propensity matching of the first page data of 752 patients with day surgery and non day surgery, 98 patients with lower extremity varicose veins, 356 patients with chronic cholecystitis and 38 patients with varicocele were finally included. Compared with non day hand, the total hospitalization cost of day surgical instruments decreased, and the cost structure changes of chronic cholecystitis, varicocele and varicose veins of lower limbs were 14.59%, 6.20% and 16.20% respectively. Among them, the general medical service fee, nursing fee and examination and laboratory fee showed a downward trend, and the fees of materials and drugs showed an upward trend. General medical service fee, nursing fee, examination and laboratory fee, clinical diagnosis fee, treatment fee, drug fee, material fee and other expenses presented a high correlation with the cost of day surgery(grey correlation>0.90). The payment method, wound healing type and discharge diagnosis can influence the cost of day surgery( P<0.05). Conclusions:Compared with non daytime surgery, the total hospitalization cost of day surgery has a certain cost control effect, but it can not reduce the cost of all projects. The main influencing factors are the internal composition of the cost, payment method and so on. The hospitals should focus on tapping the internal cost control potential of day surgery and further expanding the coverage of day surgery diseases.

ABSTRACT

OBJECTIVE:To establis h the UPLC fingerprint of Poria co cos aqueous extract ,and to investigate its relationship with sedative and hypnotic effect. METHODS :Ten batches of P. cocos from different areas were extracted with water to obtain the aqueous extract. UPLC method was adopted. The determination was performed on Waters HSS-C 18 column with mobile phase consisted of 0.1% phosphoric acid solution-acetonitrile-methanol (gradient elution ) at the flow rate of 0.4-0.2 mL/min. The detection wavelengths were set at 210 and 242 nm. The column temperature was 40 ℃,and sample size was 2 μL. The fingerprints of 10 batches of P. cocos aqueous extracts were established by using the Similarity Evaluation System of TCM Fingerprint (2012A version),and the common peaks were identified. The sedative and hypnotic effects of 10 batches of P. cocos aqueous extracts from different areas under the synergistic action of pentobarbital sodium were investigated by taking the sleeping rate ,sleep latency and sleep duration of mice as the single efficacy index. After data transformation of single efficacy index and total efficacy (single indexes calculated by analytic hierarchy process ),grey correlation analysis was used to analyze the correlation between the common peaks in fingerprint of P. cocos aqueous extract and the single efficacy index and total efficacy. RESULTS :There were 24 common peaks in 10 batches of aqueous extract of P. cocos , and 11 components were identified , i.e. 16 α-hydroxydehydrotrametenolic acid (peak 6),16α-hydroxytrametendic acid (peak 7),poricoic acid B (peak 9),dehydrotumulosic acid(peak 10),poricoic acid A (peak 12),polyporenic acid C (peak 15),3-O-acetyl-16α-hydroxydehydrotrametenolic acid (peak 17),dehydropachymic acid (peak 20),pachymic acid (peak 21),dehydrotrametenolic acid (peak 22),dehydroeburicoic acid (peak 24). Grey correlation analysis showed ,the correlation between 24 peaks and sleep duration was greater than 0.6(0.611 5- 0.811 8);the correlation between 24 peaks and sleep latency was greater than 0.6(0.605 9-0.790 4),except for peaks 14,24 and 2;the correlation of 24 peaks between sleeping rate was greater than 0.6(0.606 4-0.721 6),except for peaks 23,19,17 and 5; the correlation of 24 peaks between total efficacy was greater than 0.6(0.619 0-0.781 2),except for peaks 2,5,19. The top 10 chromatographic peaks related to the total efficacy were peak 15(polyporenic acid C ),peak 16,peak 8,peak 11,peak 12 (poricoic acid A ), peak 1, peak 7 (16 α-hydroxytrametendicacid), peak 3, peak 9 (poricoic acid B ) and peak 20 (dehydropachymic acid ). CONCLUSIONS :UPLC fingerprint of P. cocos aqueous extract was established and 11 components were identified. Ten components such as polyporus acid C are closely related to the total efficacy of sedation and hypnosis ,which preliminarily reveal the material basis of the sedative and hypnotic effect of P. cocos .

ABSTRACT

Objective:To construct the forewarning model of drug shortage in Shanxi province, so as to realize the early prediction of drug shortage.Methods:According to the drug shortage of data of each monitoring station in Shanxi province, from the four dimensions of drug factors, policy attributes, supplier factors and demander factors, 14 factors were selected for grey correlation analysis, for example whether they were urgent drugs for women and children, shortage types, etc. The main factors affecting the degree of drug shortage were selected, and the early warning analysis model based on two-step cluster analysis method was established.Results:A total of six factors with the highest correlation with the degree of drug shortage were determined in this study, in order: whether they were urgent drugs for women and children, shortage types, low-price drugs, the number of moving annual total monitoring enterprises in sale in the current month, shortage reasons, and whether they were basic drugs. Based on the two-step cluster analysis, a model of drug shortage forewarning in Shanxi Province was established, which was divided into four optimal clusters and the warning level was determined.Conclusions:This study establishes the early warning model of drug shortage in Shanxi province, which is helpful to find the risk of drug shortage as soon as possible, objectively predict the risk level, and assist the drug regulatory department to realize layered implementation and collaborative response.

ABSTRACT

Objective • To understand the structural changes and related reasons of the average cost of hospitalization of childhood pneumonia in Shanghai, and to propound related policy suggestion. Methods • New grey correlation and degree of structural variation analysis were used to research the average cost of hospitalization of 22 543 children with pneumonia from 2015 to 2018 in a children's specialized hospital in Shanghai. The average cost included 5 categories, i.e. medicine cost, material cost, examination and laboratory cost, labour cost and bed occupation cost. The relational degree and structural changes between each average cost and the average cost of hospitalization were discussed. Results • The new grey correlation analysis showed that the relational degree sort order of each average cost from the highest to the lowest were average examination and laboratory cost (γ3=1.000 0), average medicine cost (γ1=0.862 5), average bed occupation cost (γ5=0.845 1), average labour cost (γ4=0.796 8) and average material cost (γ2=0.786 3). The degree of structure variation analysis showed that the contribution rate of structure variation (CSV) sort order of each average cost in 2015-2018 from the highest to the lowest were average medicine cost (CSV1=36.22%), average bed occupation cost (CSV5=27.65%), average examination and laboratory cost (CSV3=13.91%), average material cost (CSV2=13.78%) and average labour cost (CSV4=8.44%). The average medicine cost and average material cost were negative variation. Conclusion • The average examination and laboratory cost and average medicine cost are the main factors that influenced the average cost of hospitalization of childhood pneumonia. The proportion of the average labour cost is relatively low. It is suggested that the cost structure like raising labour charges and optimizing diagnosis and treatment process should be further adjusted in order to control the growth of medical costs.

ABSTRACT

Objective:To analyze internal composition of hospitalization expense for patients with cerebral infarction. Methods:Data of hospitalization expenses from patients with cerebral infarction discharged from 2014 to 2018 were collected, and analyzed with grey correlation and degree of structural variation. Results:The costs for rehabilitation treatment, medicines and consumables mainly contributed to structural variation, accumulated to 74.63%. The costs for rehabilitation treatment (0.982), medicine (0.979), imaging (0.978) and laboratory tests (0.972) were the most relative to the average expense of hospitalization. Conclusion:It is preliminarily successful for cost control, and needs further impove to respond the contribution of professionals.

ABSTRACT

Objective: To establish a method for determination of five saponins in Panax notginseng by HPLC and comprehensively evaluate the quality of it by using grey correlation analysis. Methods: The content of notoginsenoside R1 and ginsenoside Rg1, Re, Rb1, Rd in the different origins and commercial grades of P. notginseng was simultaneously determined by HPLC, and the entire quality evaluation model was established by grey correlation analysis. Results: The established method was applied to quantify five major bioactive components in P. notginseng simultaneously with satisfactory results. Gray correlation method can distinguish the samples from genuine producing areas, qualified samples and unqualified samples, and provide reference for quality evaluation of P. notoginseng and quality evaluation of multi-index components of Chinese materia medica. Conclusion: This HPLC method was simple, accurate, stable and rapid with better separation effect, which was suitable for determination of notoginsenoside R1 and ginsenosides Rg1, Re, Rb1, Rd; The grey recognition analysis was suitable for the comprehensive quality evaluation of multi-component samples of Chinese materia medica.

ABSTRACT

Objective To study the relationship between the quality of Astragalus membranaceus and soil in Gansu Minle. Methods we sampled the medicinal materials and soils of Astragalus membranaceus in Minle County, and determined the medicinal quality, soil factors and heavy metal elements by high performance liquid chromatography, ultraviolet spectrophotometry, atomic absorption spectrophotometry, and etc. Results The quality of Minle Astragalus meets the requirements of the Pharmacopoeia standards and is low on the risk of heavy metal pollution. There was a significant negative correlation between the extract and the available potassium. There was a significant positive correlation between the astragaloside IV and the available potassium. There was a highly significant positive correlation between calycosin-7-glucoside and soluble salt. There was a significant positive correlation with available potassium, and a significant negative correlation with pH. Among them, soil pH value is the most important factor affecting the content of astragalus extract, astragaloside IV and verrucous glucoside, followed by total phosphorus, organic matter and total nitrogen. Conclusion The results revealed that the soil factors is the main influencing factor of the quality of Astragalus membranaceus in Minle County, which can provide a theoretical basis for the standardized cultivation of Astragalus membranaceus in Minle County.

ABSTRACT

Objective::To study the spectrum-effect relationship between HPLC fingerprints and anti-hepatoma activity of Paris polyphylla var. yunnanensis, P. forrestii and P. vietnamensis, and to elucidate its effective substance. Method::HPLC was used to establish the fingerprint of three extracts from the plant. The mobile phase was consisted of acetonitrile (A)-water (B) for gradient elution (0-10 min, 20%A; 10-20 min, 20%-25%A; 20-30 min, 25%-30%A; 30-40 min, 30%-35%A; 40-50 min, 35%-40%A; 50-60 min, 40%A; 60-75 min, 40%-45%A; 75-80 min, 45%-60%A), and the flow rate was 0.9 mL·min-1. The UV detection wavelength was 203 nm. Thiazolyl blue tetrazolium bromide (MTT) array was used to detect the inhibitory effects of three extracts on the proliferation of HepG2 cells, and the half inhibitory concentration (IC50) was calculated. Cluster analysis and grey relational analysis were used to analyze the data of spectrum and efficacy, and to find out the components that contributed a lot to the anti-liver cancer effect. Result::A total of 11 common peaks were identified as common peaks among HPLC fingerprints of three kinds of Paris. After treated 72 h, P. forrestii has the highest inhibitory effect on the HepG2 cells, the IC50 of P. forrestii, P. polyphylla var. yunnanensis and P. vietnamensis were 148.33, 178.87, 208.09 mg·L-1, respectively. According to the grey relational analysis, the common peaks 1-10 from P. polyphylla var. yunnanensis had great correlation to anti-tumor effect, and the common peaks 1-7 for P. forrestii, the common peaks 1-4, 6-10, N1 for P. vietnamensis, all the correlation degrees with IC50 were >0.7.Cluster analysis of variables in each Paris showed that peaks with correlation degree >0.7 could cluster with IC50. Conclusion::The established HPLC fingerprint method is reliable with good reproducibility. The peaks 1-4, 6 and 7 from three kinds of Paris have the greatest contribution to the anti-hepatoma effect.

ABSTRACT

Objective::To study the quality of Curcumae Radix from different areas, in order to screen out excellent provenances to improve the overall quality and stability of Curcumae Radix. Method::The volatile oil, alcohol extract, germacrone, germacr-1(10)-ene-5, 8-dione and curcumin were selected as comprehensive evaluation indicators to determine the content of five main components in 50 samples of Curcumae Radix from different areas. Cluster analysis was made for Curcumae Radix from different areas of Guangxi, and the fusion models of GRA(grey correlation degree) and TOPSIS (technique for order preference by similarity to ideal solution) of Curcumae Radix were established. Result::The results of cluster analysis indicated that 50 samples from different areas could be divided into four categories. The fusion models of GRA and TOPSIS were based on relative closeness γi as the index to comprehensively evaluated the quality of Curcumae Radix. Samples No. YL-2, YL-5, YL-1, YL-4 were screened out as excellent samples. The average relative closeness γi was ranked as follows: Yulin>Pingnan>Hengxian>Xingye>Zhaoqing>Qingtang>Zhongshan>Qinzhou>Guigang>Lingshan. Conclusion::The multiple attribute decision making analysis gray correlation degree and TOPSIS fusion models are used to optimize the quality evaluation of Curcumae Radix. The method is simple, objective and comprehensive, which can be promoted and provide a reference for the screening of high-quality provenance.

ABSTRACT

Objective:To investigate the spectrum-effect relationship of effect of material components in Shaoyao Gancaotang on the levels of malondialdehyde (MDA), superoxide dismutase (SOD) and adenosine triphosphatase (ATPase) in brain tissue of epileptic mice, and to reveal the material basis of the antiepileptic effect of Shaoyao Gancaotang. Method:HPLC was used to establish the fingerprint of 15 batches of Shaoyao Gancaotang, the mobile phase was consisted of acetonitrile (A)-0.1% phosphoric acid aqueous solution (B) for gradient elution (0-15 min, 19%A; 15-45 min, 19%A-50%A; 45-46 min, 50%A-90%A; 46-48 min, 90%A), the flow rate was 1.0 mL·min-1, and the detection wavelength was 237 nm. The mouse epilepsy model was induced by penicillin, the protective effect of 15 batches of Shaoyao Gancaotang on MDA, SOD and ATPase levels in brain tissue of epileptic mice was investigated. Grey correlation analysis was used to analyze the correlation between the peak areas of characteristic peaks in the fingerprint of 15 batches of Shaoyao Gancaotang and three pharmacodynamic indexes (the activities of MDA, SOD and ATPase), and the mathematical model of spectrum-effect relationship between different material components and pharmacodynamic indexes was established. Result:Shaoyao Gancaotang could increase the contents of ATPase and SOD and reduce the content of MDA in the brain tissue of epileptic mice, and most of the differences were significant (P<0.05, P<0.01). There were 28 common peaks in the fingerprint of 15 batches of Shaoyao Gancaotang, and 15 of them were characteristic peaks. Grey correlation analysis found that the characteristic peaks contributed to the activities of MDA, SOD and ATPase included peak 2 (albiflorin), peak 3 (paeoniflorin), peak 5 (liquiritin), and so on, the specific ranking was peak 3>peak 6>peak 12>peak 8>peak 2>peak 5>peak 9>peak 4>peak 10>peak 7>peak 13>peak 11>peak 15>peak 1>peak 14. Conclusion:Shaoyao Gancaotang can affect the activities of SOD, MDA and ATPase in brain tissue of epileptic mice by multi-component synergy. Simultaneously, introducing the grey correlation analysis into the correlation evaluation of the spectrum-effect relationship between components and efficacy of Chinese herbal compounds, it can objectively reflect the essence of the synergistic action of multiple components in traditional Chinese medicine (TCM), and it is an effective analysis method for screening and predicting the pharmacodynamic components of TCM.

ABSTRACT

Objective::To establish the evaluation method of Pinelliae Rhizoma (PR) resources by the analytic hierarchy process (AHP) and grey correlation degree (GCD) method, so as to explore the differences in quality components, production efficiency and appearance traits of different germplasm resources cultivated in the same environment. Method::The quality component index, efficiency index, and appearance traits index of 15 germplasm resources were measured, including moisture, total ash, extractives, total acid content, harvest index, yield, drying rate, commodity rate, decay rate and deformability of tubers. Based on AHP and grey correlation method, each indicator data was processed in a comprehensive way, its comprehensive correlation value was calculated, and different PR germplasm resources were comprehensively evaluated. Result::Based on three factors-quality composition, production efficiency and appearance traits, the comprehensive correlation value of A6 was the highest, reaching 0.749 4, which was followed by A14, A15, A7, and their comprehensive correlation values were 0.736 6, 0.726 2, 0.718 2, respectively. Therefore, the source of A6 could be used as an excellent provenance introduced to the cultivation of PR, and the provenance of A14, A15, and A7 could be used as a useful supplement. Conclusion::The method of AHP and GCD-based multi-index comprehensive evaluation is simple and comprehensive to evaluate the diversity of different PR germplasm resources, and could provide a reference for the development and utilization of resources and the screening of high-quality provenances.

ABSTRACT

Objective: To investigate the correlation between the quality and climatic factors for Gentiana officinalis H. Smith from different producing area in Gansu province, China by grey correlation degree analysis combined with CRITIC-AHP weight ed evaluation. Methods: Contents of the total secoiridoid glycosides, total alcohol extracts and the metallic elements Ca, Fe and Zn were determined for the medicinal herb G. officinalis samples from 10 producing areas in Gansu, and their quality was estimated by the CRITIC-AHP weighted evaluation. The relationship between the quality and related climatic factors was then investigated by the grey correlation degree analysis. The climatic factors included the altitude, annual mean temperature, monthly mean temperature of Janu ary and July, annual mean relative humidity and the annual mean rainfall precipitation. Results : The content of the determined com ponents showed variation in the G. officinalis samples from the 10 different producing area. The altitude, annual mean temperature, monthly mean temperature of January and July, annual mean relative humidity and the annual mean rainfall all showed certain influ ence on the G. officinalis quality, and the biggest influence was the monthly mean temperature of July. Conclusion: The present re sults of the grey correlation degree analysis identifies the main climatic factors influencing the G. officinalis quality, which provides an experimental basis for the proper area selection and planting standardization for the G. officinalis production.

ABSTRACT

To screen the antithrombotic effective components group of Trichosanthes extract, and to verify its pharmacodynamics and analyze its mechanism, the HPLC fingerprint of Trichosanthes extract (0.09, 0.45, 0.9 g·kg-1) was established, and the pharmacodynamic indexes of antithrombosis in rats with aspirin (0.01 g·kg-1) as positive control group were determined (the animals used in this experiment were approved by the Medical Ethics Committee of Wannan Medical College). The antithrombotic spectrum-activity relationship of Trichosanthes extract was studied and the effective antithrombotic ingredients group was screened by grey relational analysis. The monomer compound mixed solution (0.006, 0.03, 0.06 g·kg-1) was prepared according to the content of each component in the active component group, and the pharmacodynamics and action mechanism were studied to verify the correctness of the spectrum-effect relationship. The correlation between the 22 components of Trichosanthes extract and antithrombotic efficacy was different and showed dose-effect relationship. Cytosine, uracil, guanine, hypoxanthine, xanthine, adenine, guanosine, and adenosine are the main antithrombotic components of Trichosanthes extract. The ratio of cytosine, uracil, guanine, hypoxanthine, xanthine, adenine, guanosine and adenosine was 3∶12∶10∶5∶2∶8∶13∶14. Compared with the model group, the thrombus dry weight of each effective components group could be effectively reduced (P<0.01 or P<0.05), but there was no significant difference between each effective components group and the Trichosanthes extract group. Compared with the model group, the TXB2 content in group (0.06 g·kg-1, 0.03 g·kg-1) could be effectively reduced (P<0.01 or P<0.05), and the content of 6-keto-PGF1α could be increased in each group (P<0.01), and the TXB2/6-keto-PGF1α tended to be normal and showed a dose-effect relationship. The effect was better than that in the Trichosanthes extract group (0.45 g·kg-1) (P<0.01). The effective ingredients group has a good antithrombotic effect, its mechanism is to inhibit platelet aggregation and improve vascular endothelial function.
